20 And when he was demanded of the Pharisees, when the kingdom of God should come, he answered them and said, The kingdom of God cometh not with observation:
21 Neither shall they say, Lo here! or, lo there! for, behold, the kingdom of God is within you.

The first step in understanding this scriptures is to have a firm concept of the definition of "Kingdom of God" as explained by the bible.
The Gospel of Matthew is the Gospel that speaks the most about this subject.
Another good source of information is the prophesy by Daniel in Da 2:44.
The statement " "does not come with striking observableness" is in reference to the fact that when it does come, it will be ignored or not observed by the majority of mankind and mainstream religion.
This is specifically talking about its introduction not its full significance, which will occur later.
Once the full meaning of what the Kingdom of God entails is understood, it becomes inconceivable to conclude that the Kingdom is within or in the heart of the Pharisees as they proved to be an enemy of Jesus who was to be King of that Kingdom, and everything the Kingdom stood for.
Rather the word "within" can be translated : amongst you, or with you. This claim Jesus could make since he was the King of the kingdom.
The term "Kingdom of God" and the time of it's arrival are both subject of sizable information that go beyond the parameters of this discussion, but are important in understanding the message of the bible.
