The Immaculate Conception does refer to the spiritually quality of Mary, not Jesus. Both this doctrine and the doctrine of Papal Infallibility were declared by Pius IX in the latter half of the nineteenth century. They are relatively very new doctrines.

If I don't believe in Muhammad's miraculous night journey to Jerusalem and then to heaven and back, does that prove I only want to destroy Islam?

The adherents of any particular religion should not assume that skepticism about their religion's claimed miracles indicates hostility. Such skepticism is perfectly reasonable, and to be expected.
